可以这样表达，另外用whom 也是可以的 但一般用who-- 按严格语法应该是whom，但是约定俗成大家都用who（不合语法，但洋鬼子就喜欢这样，我们也没办法）
更好的说法：
Which one do you like most, your brother or your sister?

most是不对的，只有两类比较项，用better

不对,whom应用在句中

whom可以这么用
如果要找语病的话，我想是在like和most之间应该加the,
就是：whom do you like the most, your brother or your sister?

who可以做主格,也可以做宾格
whom只能做宾格
用who肯定没错,但是用whom更准确.(根据上下文意思)

应该把most改为than

首先这个句子用most是不对的,这样就成了*表达,而后面可供选择的只有your brother和your sister两者,因此应该用比较级:Who do you like better,your brother or your sister?[注]这句话开头疑问词用whom未尝不可...

Whom 用得没错，因为它指待的是宾语，当然你也可以用who。Who是主语宾语都可以用的。
不过这个句子是错的，因为后面只是给了brother和sister你选择，也就是说是两个选择，两者之间的比较的话不能用*，只能用比较级。就是说你只能用more，不能用most，当然你也可以用better。
所以应该改成：Whom do you like more, your brother or your sister? 或者：whom do you like better，your brother or your sister?或者 Who do you like more, Who do you like better.

其实WHO 何whom 没有什么太大的区别， 这句话是对的。
